Hubert Vincent Clairborn Holcombe was born 5 January 1828 in West Granville, Massachusetts. He was the son of Dr. Vincent Holcomb. Hubert V. C. Holcomb first began to study medicine with his father; he also studied in Lee, Massachusetts, and he graduated at Castleton, Vermont. He worked in practice with his father in Granville until 1854, when he moved to Branford, Connecticut.

Dr. Hubert V. C. Holcombe enlisted on 14 August 1862, serving during the Civil War as Surgeon of the 15th Regiment of the Connecticut Volunteer Infantry. He was discharged in New Bern, North Carolina, on 27 June 1865. Dr. Holcombe returned to his practice in Branford after the war, where he died in 1878.
